Qualitative test for carbotiydrates.

  1. Molisch’s Test  2 ml of sample +2 drops of molisch’s reagent + 1 ml conc. H 2 So 4 along sides of test tube. Observation – Purple ring at junction of two layers. Principle – It is general test for all carbohydrates. Conc. H 2 So 4 hydrolysis glycosidic bond to yield monosaccharides that in the presence of acid get dehydrated to form furfural or its derivatives. Furfural derivatives reacts with sulphonated -napthol sol, (5% W/V in ethanol = prepared fresh as it is unstable) to form purple complex.
  2. Fehling’S Test  1 ml of sample + 1 ml of fehling’s A sol. + 1 ml of fehling’s B sol. -Boil. Observation – Brick-red PPt. Principle – This test is for reducing sugars i.e. sugar containing free aldehyde or ketonic group. Fehling’s A – CuSo 4 sol. Fehling’s B – Sol. Of KOH + Na-K tartarate(-rochelle’s salt) Reducing sugar reduce CuO (+2) of fehling’s reagent to Cu 2 O(+1) which forms red PPt. The free =C=O group of sugar is oxidized to -CooH group. Rochelle’s salt act as chelating agent.

  1. Jodide test  1ml sample + 5 drops of I 2 sol (in KI) Observation – Violet colour. Principle – I 2 forms coloured adsorption complex with polysaccharides. Starch gives blue colour while glycogen given reddish brown colour.
