Back to Home Page KB02 Basics of Microscopy Title: Introduction to Microscopy Objective: To become familiar with the basic components of a microscope, it, and how to go about using it to visualize samples, and how to load a sample for viewing. Procedure:  Review parts and components of a microscope.  Load sample slide onto microscope to be viewed.  Choose a magnification.  Adjust the microscope for a clear view of a given sample. Notes: Parts of Microscope:

  • Eyepieces (ocular lens): the lenses we look through; pull apart; use both eyes to see one clear image. Adjust for comfort if needed.
  • Arm/neck: The part we hold when carrying the microscope; support with one hand under the base; grab when moving.
  • Objective lens: Lenses that magnify the sample; the shortest one gives the least magnification; the longest gives the most.
  • Revolving nosepiece: The part that holds and rotates the objective lenses.
  • Stage: Flat area where we place our slides to view the samples; use a clamp on the coverslip of the slide.
  • Stage controls: Knobs that move the stage to position the sample under the lenses.
  • Stage clips: Hold the slide in place on the stage.
  • Focus knob (coarse/ fine focus): Located on the side; the big knob makes large adjustments, the small knob fine-tunes for sharp detail.
  • Iris diaphragm: Controls how much light passes through the sample, located below the stage.
